Soul segue la storia di Joe Gardner (voce originale Jamie Foxx), insegnante di musica in una scuola media, che desidera suonare nel famoso jazz club di New York The Blue Note. Joe perderà la vita mettendo involontariamente il piede sbagliato in una grata fognaria aperta, ma la morte non rappresenterà la fine del suo viaggio. Quando l'anima di Joe lascerà il suo corpo, inizierà un nuovo sorprendente viaggio, che porterà la sua anima in un regno cosmico, il Seminario "You", dove vengono create e perfezionate le anime. Nel Seminario "You", le anime vengono infatti istruite e solo dopo essersi diplomate hanno la possibilità di entrare nel corpo delle persone. Sarà proprio qui che l'anima di Joe si perderà e grazie all'aiuto di tutte le altre anime, viaggerà nel cosmo per ritrovare la strada verso la Terra.
